MORPHOLOGICAL CHANGES IN THE COLON AND COMPOSITION OF PERIPHERAL BLOOD LYMPHOCYTES IN ACUTE COLITIS IN MICE WITH DIFFERENT RESISTANCE TO HYPOXIA

Introduction. According to literature, there is individual and linear variability of severity of acute ulcerative colitis induced by dextran sodium sulfate in mice. The resistance to hypoxia could be one of the possible mechanisms underlying the individual differences in the severity of acute colitis. There is no data about features of the inflammatory process during the acute ulcerative colitis in mice with different hypoxia tolerance. The aim of the study. To characterize the morphological changes in the colon and the composition of peripheral blood lymphocytes in acute ulcerative colitis C57Bl/6 mice with different resistance to hypoxia. Methods. Resistance to hypoxia was determined using the period of a lifetime at the height of 10,000 m, that corresponds to the interval between the time of lifting in the hypobaric chamber before taking the lateral position. One month later acute ulcerative colitis was modeled by replacing drinking water with the aqueous solution of 1,5% dextran sodium sulfate. Results. Susceptible to hypoxia mice are characterized by more pronounced clinical symptoms, a greater prevalence of ulcers with fibrin in the medial and ulcers without epithelization in the proximal part of the colon than mice than tolerant mice. In susceptible animals, there is a marked decrease in lymphocytes, absolute number of T-lymphocytes (CD3+), Т-helpers (CD3+CD4+), cytotoxic Т-lymphocytes (CD3+CD8+), Т-regulatory cells (CD4+CD25+FOXP3+), absolute and relative number of B-lymphocytes (CD19+) in the peripheral blood, while in the tolerant mice no changes are observed. Conclusion. Variations in the severity of acute colitis in animals differing in resistance to hypoxia were revealed: susceptible mice are characterized by more pronounced clinical symptoms, a greater prevalence of ulcers with fibrin in the medial and ulcers without epithelization in the proximal colon, marked decrease in all subsets of lymphocytes in the peripheral blood.
