Введение. Высвобождение медиаторов из тучных клеток или базофилов является ключевым событием для множества острых аллергических и воспалительных процессов. Клеточная линия базофильной лейкемии крысы RBL-2H3 широко используется в качестве модели для изучения дегрануляции тучных клеток. Цель исследования. Отработка условий (подбор оптимального индуктора, времени экпозиции и концентраций) индукции дегрануляции клеточной линии базофильной лейкемии крысы RBL-2H3. Материал и методы. Клетки стимулировали известными индукторами: веществом 48/80 и кальциевым ионофором А23187 в широком диапазоне концентраций. Проводили инкубацию клеток с индукторами 10, 25 и 45 мин. Дегрануляцию оценивали по активности фермента β-гексозаминидазы в супернатантах и клеточных лизатах. Результаты. Наблюдали различия в степени дегрануляции в зависимости от концентрации и времени инкубации с тестируемыми индукторами. Зависимость от концентрации имела характерную колоколообразную форму для обоих веществ. Заключение. В целом индукция дегрануляции кальциевым ионофором А23187 была более эффективной и приводила к более высокому (по сравнению с веществом 48/80) проценту чистого выброса фермента β-гексозаминидазы. Обсуждены механизмы действия вещества 48/80 и ионофора А23187 в качестве индуктора дегрануляции тучных клеток.
