THE ROLE OF P53 AND NOXA IN THE PATHOGENESIS OF EARLY MISCARRIAGES

Introduction. Missed and spontaneous abortions are one of the most important causes of the reproductive loss. The aim of the study: to study specialties of mRNA expression of p53 and proteins of its signaling pathway in the decidual tissue at 6–10 weeks of gestation in patients with missed and spontaneous abortions. Methods. mRNA expression of p53, p63, p21 (CDKN1A), BCL-2, Bax, Noxa and MDM2 in the decidual tissue (endometrium) in 50 patients with missed abortions, 47 patients with spontaneous abortions and 57 patients with artificial abortion (control group) at 6–10 weeks of gestation was studied using the reverse transcription quantitative polymerase chain reaction (RT-qPCR). Results. mRNA expression of p53 and its homolog p63 had no significant differences in patients with missed and spontaneous abortions compared with the control group. The significant increase of pro-apoptotic protein Noxa, accompanied with the elevation in anti-apoptotic protein BCL-2 was observed in the decidual tissue of patients with missed abortions. In patients with spontaneous abortions, the increase of Noxa only has been observed. No significant changes of p21 mRNA were discovered in both groups, which indicates, that the arrest of the cell cycle is not involved in the pathogenesis of early miscarriages. No significant changes of both Bax and MDM2 mRNA expression were discovered in the decidual tissue of patients with missed and spontaneous abortions. Conclusion. The increase of Noxa mRNA expression in the decidual tissue of patients with missed and spontaneous abortions can be suggested to be potent of playing an important role in the pathogenesis of these complications. The significant increase of BCL-2 in patients with missed abortions, when embryo resorption has happened, probably protects nearby embryos in case of the multiple pregnancy.
