THE ROLE OF OXIDATIVE PROTEIN MODIFICATION IN REDOX-DEPENDENT REGULATION OF TUMOR CELL APOPTOSIS

Introduction. According to modern understanding, oxidative stress along with apoptosis dysregulation plays an important role in the pathogenetic mechanisms of tumor growth; formation of oxidative stress is accompanied by oxidative modification of cell protein molecules. The research objective. To determine the role of oxidative protein modification in redox regulation of Jukart tumor cell apoptosis. Material and methods. Jukart tumor cells have been used in the research (T-lymphoblastic leukemia). The amount of annexin-positive and annexin-propidium iodide-positive cells, the concentrations of protein-SH groups, protein-bound gluthatione, carbonyl protein derivatives, bitirozine and oxidated tryptophan have been determined. Results. In experiments in vitro peculiarities of the participation of oxidative protein modification in realization of programmed Jukart cell death have been identified under the conditions of cultivation with the blocking agent of protein-SH groups in the concentration 5 mM – N-ethylmaleimide, protector of protein-SH groups in the concentration 5 mM – 1,4-dithioerythritol and/or apoptosis inductor in the concentration 10 mkM – dexamethasone. Conclusion. It has been shown that activation of the thanatogenic Jukart tumor cell programme under the effect of 1,4 – dithioerythritol was accompanied by a rise in the concentration of protein-SH groups against the backdrop of a decrease in the amount of oxidated tryptophan and bitirozine; in the presence of N-ethylmaleimide it was accompanied by accumulation of carbonyl gluthatione derivatives.
