RELATIONSHIP OF PLOIDITY AND LOCAL IMMUNITY IN TUMORS OF THE COLON

Introduction. The biologic behaviour of the tumor can be determined by both its own characteristics including ploidity of the tumor cells and peculiarities of its lymphocytic and cytokine microenvironment, in particular. The aim of the study. To perform a comparative study of lymphocytic and cytokine environment of diploid and aneuploid malignant tumors of colon. Methods. DNA-cytometry (CycleTESTTMPLUS DNA Reagent Kit) and subsets of lymphocyte amount (T-B-NK) were studied by flow cytometry with the use of cytofluorometer BD FACSCantoII (BD, USA). ELISA test was used for assessment of cytokines levels in tissue samples.The study was performed in tissue samples of colon cancer, peritumoral area and resection line. Results. Some differences in the subsets pattern of lymphocytes were revealed, mostly pronounced in peritumoral area: aneuploid tumors contained more CD3+ and CD3+CD4+ and less CD19+ cells than diploid tumors. The pattern of lymphocytes subsets in aneuploid tumor samples was similar to their blood pattern that indicates rather their peripheral origin than mucosal one. When the amount of aneuploid tumor cells exceeded 70%, a higher amount of CD3+CD4+ and lower one of CD3+CD8+ lymphocytes were noted both in tumor and peritumoral area. In the tissue samples of diploid tumors and their peritumoral area a higher level of IL2 was observed than in aneuploid ones, but levels of IL1, IL6, IL8, TNF-α in tumor samples exceeded their levels in non-malignant tissues independently of the ploidity of the tumor. Conclusion. Factor of tumors` malignancy (aneuploidity) and some indices of local immunity seem to be interrelated.
