THE STUDY OF INTERFERON GENE EXPRESSION IN THE HUMAN FIBROBLAST CULTURE IN RESPONSE TO IMMUNOMODULATORS AND VIRAL INFECTION

Introduction. Diploid human fibroblasts lines (DHFL) are widely used in diagnostic virology and experimental studies. One of the directions of use DHFL in virology and immunology is to create on their basis of a test system for determining the activity of interferon (IFN). The aim of the study. Study spontaneous and inducible interferon (IFN) gene expression in the diploid human fibroblast lines (DHFL) in situ, and in response to 2 immunomodulators and vesicular stomatitis virus (VSV). Results. The data obtained show that DHFL are a good substrate for VSV propagation, which justifies their use for serum IFN testing. In the present study we characterized IFNα/β, γ and chemokine IP-10 gene expression in DHFL in normal conditions, and in response to stimulation with 2 immunomodulators possessing antiviral and interferonogenic activity. We show that DHFL were able to spontaneously produce chemokine IP-10 mRNA induced by IFNγ. Expression of IP-10 significantly increased in response to exogenous IFN-α and LPS. Conclusion. The data obtained indicate that DHFL can be used as a test system for studying the mechanisms of action of immunomodulators and anti-viral drugs.
