THE STUDY OF TOXICOLOGICAL PROPERTIES OF THE POLYMERIC FORM OF AN ETOPOSIDE

Introduction. The clinical use of the antitumor drug etoposide is associated with many side effects, limiting its application in the clinical practice. One of the ways to reduce the toxicity and increase the effectiveness of the action of etoposide can be its application in the composition of particles based on biodegradable polymers. The aim of the study. The investigation of toxic properties of etoposide in the form of particles of submicron size based on poly(lactide-co-glycolide). Мethods. A comparative toxicological study of the developed form of etoposide and the commercial drug «Etoposide-Ebewe» was carried out on healthy BALB/c mice to determine the acute and course toxicity. Results. The LD50 value of the developed form of etoposide as shown to be approximately 2 times higher in comparison with the commercial reference preparation for the intravenous and intraperitoneal administration. The study of the tolerability of the course administration of drugs, assessed by lethality, the general condition of animals, behavioral reactions, the clinical picture of intoxication during and after the course of the administration, the dynamics of body weight of animals, hematological studies also indicate that the toxic effect of the delivered form of etoposide is less pronouncedif compared to the reference preparation. Conclusion. The developed form of etoposide shows less pronounced toxicity in comparison with a commercial reference drug.
