THE EXPRESSIVE METHOD OF DNA ISOLATION FROM FROZEN BLOOD

Introduction. Mass genotyping, aimed both at identifying of genetic characteristics of populations and the creation of genomic libraries, scientific research, was implemented on large samples, required the accumulation and/or conservation of samples of the biomaterial as a source of genomic DNA. The known methods of DNA isolation from frozen blood are laborious and time- consuming. Aim. A search for a new method for genomic DNA isolating from the frozen blood. Material and method. For DNA isolation and subsequent genotyping using the allele-specific polymerase chain reaction (PCR) with electrophoretic detection of the results, samples of frozen blood were stabilized by EDTA collected under expedition conditions and stored at -25°C for several years were used, as well as samples of freshly collected blood samples, some of which were frozen and stored at -25°C, and some were used immediately after blood sampling for DNA isolation. The DNA isolation was carried out with the help of commercial reagent «DNA-EXPRESS-blood» manufactured by Lytech Co. Ltd. (Moscow). Results. The use of the new method of genomic DNA isolating from frozen blood makes it possible to obtain a sufficient amount of DNA from long-term frozen blood for PCR. The procedure for the isolation of DNA was shortened in duration and in the number of stages compared with the commercial method. The parallel isolation of DNA from samples of frozen blood by the proposed method and from freshly collected bloodby a commercial method allow obtaining the same result in genotyping. Сonclusion. The new express method for the isolationof genomic DNA from frozen blood is presented.
